Abstract:—The source for these markers was, on the one hand, a public 'expressed sequence tag' (EST) database from Ananas comosus, and on the other hand, the three bromeliads Fosterella rusbyi, Dyckia marnier-lapostollei and Deuterocohnia longipetala were partially sequenced using the 454 technique. Both approaches provided a total of 164,137 raw sequences, on the basis of which several thousand perfect microsatellites were identified and numerous flanking PCR primer pairs were derived for both the nuclear and the chloroplast genome. After extensive functionality tests with a focus on interspecific transferability and intraspecific variability of the microsatellite loci in the respective target species, 48 nuclear and seven plastid microsatellite markers were finally established.
